FSK3533 New Super-resolution, Light-sheet, STED-FCS and FRET-FCS methods at Scilifelab 3.0 credits

Content and learning outcomes

Course disposition

Lab-exercises and discussion groups will be web-based. If the situation is improved the course will be given at the 3rd floor in the Gamma building at Scilifelab Solna. 

The techniques covered are MoNaLISA which is a new fluorescence super-resolution technique, Light-sheet and lattice light-sheet imaging, FCS combined with STED­microscopy to generate a nanoscopic detection focus, and FCS combined with FRET for enhanced sensitivity to detect rare protein oligomers. 

Each of the four techniques will be covered in two theoretical lectures and two hands­on laboratory exercises. The students will also at four occasions participate in smaller discussion groups, where each occasion focuses on one of the four techniques. The lectures, laboratory exercises and discussion sessions will be given during week 1. The second week will be devoted to litterature studies and the examination, which will be in the form of formulating a project plan involving one of the techniques.

  • Introduction to fluorescence spectroscopy and imaging
  • Principles of fluorescence super-resolution microscopy
  • STED- and RESOLFT-based super-resolution microscopy
  • The MoNaLISA imaging technique.
  • Light-sheet imaging
  • Lattice light-sheet imaging
  • Introduction to Fluorescence Correlation Spectroscopy (FCS)
  • Combination of the super-resolution technique STED with FCS
  • Combination of Förster Resonance Energy Transfer (FRET) and FCS

Intended learning outcomes

  • To understand the theory and methology behind the techniques MoNaLISA, Light­sheet and lattice light-sheet imaging, and FCS combined with STED and FRET.
  • To have hands-on experience of using these techniques on relevant biological samples.

Specific prerequisites

M. Sc. in Physics or equivalent

Recommended prerequisites

Solid background in Physics, Chemistry and Biology. Experience working with complex laboratory equipment.

Equipment

All necessary equipment is available at the Advanced Light Microscopy Facility {ALM) at Gamma 3, Scilifelab. 

Literature

Various articles, lecture notes, and training manuals, made available to participating students. 

Examination and completion

If the course is discontinued, students may request to be examined during the following two academic years.
